Berkshire open their defence of the Unicorns Trophy when they play Cornwall at The Oratory School, Woodcote, on Sunday (11am).
Unlike previous seasons when there were four group games, this 50/50 competition is now a straightforward knock-out event.
The Unicorns Championship and Trophy winners will be forced to make a number of changes to the team that defeated Lincolnshire in the one-day final at Wormsley last August.
Waqas Hussain is still recovering from the serious facial injuries he suffered during a freak training accident last month, while fellow opening bat Ollie Wilkin is taking a break from cricket.
In their absence, Chris Peploe and Andy Rishton are set to open the innings, as they had in Berkshire's four T20 games two weeks ago, when they defeated Shropshire twice at Wargrave and won Buckinghamshire once at Amersham, leaving them top of their group.
On Tuesday, Rishton helped Loughborough MCCU defeat Australian Universities by 101 runs. He made 86 off 79 balls (1x6, 12x4) in the hosts' 246 all out but didn't take a wicket (0-30) as the tourists were dismissed for 145.
This is a second round tie, with Berkshire having received a bye in the opening round while Cornwall defeated Dorset by 7 wickets at North Perrott with Henley's Matt Rowe making 53 not out in his side's 156-3.
The Oratory ground is said to be "looking in immaculate condition".
