They bowled out their visitors for 100 in 36.5 overs with Lauren Bell taking 3-21 off 8.5 overs, Anya Shrubsole 2-9 off 5, Alice MacLeod 2-8 off 6 and Kate Coppack 1-15 off 6. The other wicket fell to a Bell run-out.
Not one Essex batter reached double figures and the visitors only reached 100 thanks to a remarkable 47 extras, which included 35 wides.
Berkshire lost openers Emily Cunningham (0) and Mille Atherton (1) to leave them 8-2 before skipper MacLeod and England allrounder Shrubsole laid the foundations for the victory.
MacLeod made 30 and Shrubsole 22. Bell failed to get off the mark but Amanda Potgieter saw her side home with an unbeaten 35 (5x4) off 56 balls, while Ellen Watson was 5 not out. Berkshire reached their target at 101-5 off 35.2 overs, so a good start for new head coach Danni Warren.
